Research Paper : The Anti-obesity Effect of Cudrania Tricuspidata Ethanol Extract by Focusing on the Changes of Hepatic and Adipose Tissue in HFD-induced Obese Mice

Abstract
In order to evaluate the anti-obesity effect of Cudrania tricuspidata Bureau ethanol extract in HFD-induced obese mice, we conducted histological and molecular biological experiments. The thirty-five, six-week-old C57BL/6 male mice were classified into five groups (7 in each group) such as normal group (N), control group (C), positive control group (PC, 0.5% catechin), experimental group 1 (E1, 2% Cudrania tricuspidata Bureau stem ethanol extract, CTSEE) and experimental group 2 (E2, 2% Cudrania tricuspidata Bureau leaves ethanol extract, CTLEE). After inducing the obesity by providing high-fat diet except normal group for four weeks, we provided catechin, CTSEE and CTLEE for additional five weeks. Daily body weight gain significantly decreased (p<0.05) in E1 group, compared to C group. In histological observation (H&E staining) of hepatic tissue, the lipid deposit of E1 and E2 group was decreased compare to C group. The size of epididymal adipose tissue cells decreased (p<0.05) significantly in E1 group, compared to C group. In antioxidant activities of hepatic tissue, there were significant increases (p<0.05) of SOD and CAT, and significant decreases (p<0.05) of XO and TBARS in PC, E1 and E2 group, compared to C group, which indicated catechin, CTSEE and CTLEE showed superior antioxidant ability. The mRNA expression related to the lipid metabolism was evaluated in hepatic tissue and epididymal adipose tissue.
