The Cell viability & Patch test of Semi-permanent Eyebrow Cosmetic depending on Various pH Levels

Abstract
This study was to evaluate the safety of semi-permanent eyebrow cosmetics depending on various pH levels. We prepared the liquid type of semi-permanent eyebrow cosmetics throughout the changing the pH; pH 5.0, pH 4.0 and pH 3.0 respectively. The samples have evaluated in human fibroblast skin cells using cell viability assay(Trypan blue exclusion test) and skin patch test(contact hypersensitivity allergy test; WKIRB-201706-038-02). The patch test was conducted on 30 female volunteers. As the results of the safety evaluation, the survival rate of fibroblast cells was decreased in the existence of acidity regulator(citric acid and sodium citrate). This effect of acidity regulator on the survival of fibroblast cells was dose-dependent and pH level(below 6.0). Whereas, there was no positive reaction from skin patch test in sample A(pH 5.0), sample B(pH 4.0). The interpretation of patch test results is very important for safety. There was no skin reaction in response to the control(pH 6.0), sample A(pH 5.0) and sample B(pH 4.0), whereas one case(3.3%) showed a doubtful reaction(+/-; erythema) to sample C(pH 3.0). As a prominent finding, the application of pH 4.0~6.0 is safe as semi-permanent eyebrow cosmetic.
