Research Paper : Study on the Protective Effect of Caffeic Acid on the Human Skin Fibroblast Damaged by Hair Bleach Agent in Culture

Abstract
To clarify the effect of caffeic acid on the cytotoxicity induced by hair bleach agent, colorimetric assay was carried out to determine the cell viability on the human skin fibroblasts (Detroit 51) exposured to various concentrations of hydrogen peroxide (H2O2) of hair bleach agent. The effect of caffeic acid on the cytotoxicity induced by H2O2 of hair bleach agent was examined by cell viability, lactate dehydrogenase (LDH) activity and the measurement of ferric thiocyanate (FTC). In this study, H2O2 of hair bleach agent decreased cell viability according to dose dependently in these cultures and XTT50 values was measured at concentrations of 40 uM of H2O2 of hair bleach agent after human skin fibroblasts were exposed to 20~40 uM of H2O2 of hair bleach agent for 7 hours, respectively. H2O2 of hair bleach agent was highly toxic on cultured human skin fibroblasts. In the effect of caffeic acid on H2O2 of hair bleach agent, caffeic acid significantly increased cell viability and decreased LDH activity and FTC in dose dependently compared with H2O2 treated group. From these results, it is suggested that H2O2 of hair bleach agent was highly toxic on cultured human skin fibroblasts, and caffeic acid of phenolic compound showed the protective effect against H2O2 of hair bleach agent.
