Abstract |
In this study, bacterial species contaminated nail care tools were analyzed to estimate contamination sources. Bacterial genomic DNA was extracted from the disinfected or the used pusher, nipper, file, sanding, finger bowl and pedispa bath in 8 to 10 nail saloons (A from J). The genomic DNA was used as a template for 16S-rDNA amplification by polymerase chain reaction (PCR) technique. 16S-rDNA was amplified from the bacterial genomic DNA obtained from the used nail care tools but was not from the disinfected ones. The 16S-rDNA variable region (16S-VR) was amplified from the 16S-rDNA by PCR. Total 15, 12, 12, 12, 16, and 22 of DNA bands were obtained by the temperature gradient gel electrophoresis (TGGE) of the 16S-VR obtained from files, pushers, sandings, nippers, finger bowls, and pedispa baths, respectively. Number of bacteria contaminated the disinfected and the used nail care tools was lower than 6 and higher than 130, respectively, per cotton swab that was used to collect bacterial cells from the nail care tools. These results show that the bacteria contaminated the nail care tools may be prevented effectively by disinfections and various bacteria can contaminate the nail care tools by only once use for customers. |
